Molecular Characterization of Quinolone Resistance Determinants in Non-Typhoidal Salmonella Strains Isolated in Tehran, Iran

Background: Quinolone resistant Salmonella serotypes have been reported in recent years and become increasingly widespread worldwide. Objectives: We evaluated the molecular mechanism of quinolone resistance non-typhoidal strains isolated from clinical samples Tehran, Iran. Methods: The present study included isolates originated hospitalized individuals outpatients Serotyping nalidixic acid-resistant was done by slide agglutination method. Then, resistance-determining region (QRDR) topoisomerase gene gyrA plasmid-mediated (PMQR) determinants were detected using polymerase chain reaction (PCR) Restriction fragment length polymorphism (RFLP) analysis also employed to determine possible mutation those strains. Mutant enzymatic digestion, their PCR products sequenced immediately. Results: Amongst 141 isolates, 60% showed acid resistance, whereas none them ciprofloxacin-resistant. commonly prevalent S. Enteritidis Infantis. Of 85 strains, 17 (20%) harbored qnrS gene. However, quinolone-resistant did not detect qnrA qnrB genes. PCR-RFLP sequencing QRDRs indicated that 16 (18.8%) had mutant patterns, most common point serine phenylalanine at position 83. Conclusions: Our results demonstrated mutations existence important mechanisms human origin. Other alternative such as alterations expression efflux pumps, should be studied provide greater insight into isolates.
